This variant lies in the PNPLA1 gene (transcript NM_001374623.1) at coding-DNA position 614, where C is replaced by T; at the protein level this means replaces proline at residue 205 with leucine — a missense variant. Submitter rationale: Variant summary: PNPLA1 c.614C>T (p.Pro205Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251434 control chromosomes. c.614C>T has been reported in the literature in at-least one individual affected with Autosomal recessive congenital ichthyosis (example: Youssefian_2018). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
