Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_139276.3(STAT3):c.1366-20A>G, citing LabCorp Variant Classification Summary - May 2015: Variant summary: STAT3 c.1366-20A>G alters a non-conserved intronic nucleotide that could alter mRNA splicing, although 5/5 computational tools predict no significant effect on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 120986 control chromosomes (ExAC). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1366-20A>G in individuals affected with Hyper IgE Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
