Truncations downstream of this position have been classified as likely pathogenic by our laboratory (e.g., c.8391_8392delTG, p.Gly2798fsX2 and c.8443delC, p.Leu2815fsX31). The variant was absent in 246190 control chromosomes (gnomAD). To our knowledge, no occurrence of c.8359_8360dupCT in individuals affected with Marfan Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
