This variant lies in the DUOX2 gene (transcript NM_001363711.2) at coding-DNA position 3116, where G is replaced by A; at the protein level this means replaces arginine at residue 1039 with glutamine — a missense variant. Submitter rationale: Variant summary: DUOX2 c.3116G>A (p.Arg1039Gln) results in a conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 251362 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3116G>A has been reported in the literature in individuals affected with Hypothyroidism (Matsuo_2016, Wang_2021). These reports do not provide unequivocal conclusions about association of the variant with Thyroid Dyshormonogenesis 6. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 27166716, 33631011). Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
