This variant lies in the SUZ12 gene (transcript NM_015355.4) at 5 bases into the intron immediately before coding-DNA position 275, where C is replaced by G. Submitter rationale: The c.275-5C>G variant in the SUZ12 gene has been observed in internal GeneDx clinical exome sequencing data in association with overgrowth, dysmorphic features, intellectual disability, advanced bone age, and cryptorchidism. In silico analysis, which includes splice predictors and evolutionary conservation, supports a deleterious effect. The c.275-5C>G variant is not observed in large population cohorts (Lek et al., 2016). Therefore, we interpret c.275-5C>G as a variant of uncertain significance.
