NM_007194.4(CHEK2):c.1250T>C (p.Leu417Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L417P variant (also known as c.1250T>C), located in coding exon 10 of the CHEK2 gene, results from a T to C substitution at nucleotide position 1250. The leucine at codon 417 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
