NM_005359.6(SMAD4):c.1258C>T (p.Arg420Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome; Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R420C variant (also known as c.1258C>T), located in coding exon 9 of the SMAD4 gene, results from a C to T substitution at nucleotide position 1258. The arginine at codon 420 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
