NM_000527.5(LDLR):c.2510A>G (p.His837Arg) was classified as Uncertain significance for Familial hypercholesterolemia by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This missense variant replaces histidine with arginine at codon 837 of the LDLR protein. This variant is also known as p.His816Arg in the mature protein. Computational prediction tools indicate that this variant has a deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in one individual affected with familial hypercholesterolemia (PMID: 29269200) and in one individual affected with myocardial infarction (PMID: 25487149). This variant has been identified in 4/251452 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
