NM_174936.4(PCSK9):c.886G>A (p.Val296Ile) was classified as Uncertain Significance for Hypercholesterolemia, autosomal dominant, 3 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the PCSK9 gene (transcript NM_174936.4) at coding-DNA position 886, where G is replaced by A; at the protein level this means replaces valine at residue 296 with isoleucine — a missense variant. Submitter rationale: Variant of Uncertain Significance due to insufficient evidence: This missense variant is located in the catalytic peptidase domain of the PCSK9 protein. Computational prediction tools and conservation analyses suggest that this variant may not impact the protein function. Computational splicing tools suggest that this variant may not impact the RNA splicing. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with familial hypercholesterolemia in the literature. This variant has been identified in 6/227772 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the pathogenicity of this variant conclusively.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
