Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.2371C>G (p.Leu791Val), citing Ambry Variant Classification Scheme 2023: The p.L791V variant (also known as c.2371C>G), located in coding exon 9 of the BRCA1 gene, results from a C to G substitution at nucleotide position 2371. The leucine at codon 791 is replaced by valine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
