Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000384.3(APOB):c.400G>A (p.Ala134Thr), citing Ambry Variant Classification Scheme 2023: The p.A134T variant (also known as c.400G>A), located in coding exon 5 of the APOB gene, results from a G to A substitution at nucleotide position 400. The alanine at codon 134 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
