NM_000465.4(BARD1):c.1262C>G (p.Ala421Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A421G variant (also known as c.1262C>G), located in coding exon 4 of the BARD1 gene, results from a C to G substitution at nucleotide position 1262. The alanine at codon 421 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
