NM_000218.3(KCNQ1):c.880G>A (p.Val294Met) was classified as Uncertain Significance for Long QT syndrome by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the KCNQ1 gene (transcript NM_000218.3) at coding-DNA position 880, where G is replaced by A; at the protein level this means replaces valine at residue 294 with methionine — a missense variant. Submitter rationale: Variant of Uncertain Significance due to insufficient evidence: This missense variant is located in the extracellular linker between transmembrane domain S5 and pore-forming region of the KCNQ1 protein. Computational prediction tools and conservation analyses suggest that this variant may not impact the protein function. Computational splicing tools suggest that this variant may not impact RNA splicing. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 4/245602 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the pathogenicity of this variant conclusively.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
