This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 1634, where A is replaced by G; at the protein level this means replaces lysine at residue 545 with arginine — a missense variant. Submitter rationale: The MSH6 c.1634A>G (p.K545R) variant has been reported in a large case-control study of breast cancer in 1/60466 cases and 0/53461 controls (PMID: 33471991). It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 629438). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
