Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002335.4(LRP5):c.1828G>A (p.Gly610Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 610 of the LRP5 protein (p.Gly610Arg). This variant is present in population databases (rs80358313, gnomAD 0.003%). This missense change has been observed in individual(s) with autosomal dominant osteoporosis with retinopathy and autosomal recessive osteoporosis-pseudoglioma syndrome (PMID: 15981244, 16252235, 20340138). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 6294). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt LRP5 protein function with a positive predictive value of 95%. Experimental studies have shown that this missense change affects LRP5 function (PMID: 16252235). For these reasons, this variant has been classified as Pathogenic.
