This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 1357, where C is replaced by G; at the protein level this means replaces proline at residue 453 with alanine — a missense variant. Submitter rationale: This sequence change replaces proline with alanine at codon 453 of the BRCA2 protein (p.Pro453Ala). The proline residue is weakly conserved and there is a small physicochemical difference between proline and alanine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
