Uncertain significance for Cardiomyopathy — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_001005242.3(PKP2):c.1379-2000T>A, citing ACMG Guidelines, 2015. This variant lies in the PKP2 gene (transcript NM_001005242.3) at 2000 bases into the intron immediately before coding-DNA position 1379, where T is replaced by A. Submitter rationale: This missense variant replaces valine with aspartic acid at codon 496 of the PKP2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with dilated cardiomyopathy (PMID: 30685992). This variant has also been identified in 11/220230 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
