Uncertain Significance for Long QT syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000238.4(KCNH2):c.2832G>C (p.Glu944Asp), citing ACMG Guidelines, 2015. This variant lies in the KCNH2 gene (transcript NM_000238.4) at coding-DNA position 2832, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 944 with aspartic acid — a missense variant. Submitter rationale: Variant of Uncertain Significance due to insufficient evidence: This missense variant is located in the cytoplasmic domain of the KCNH2 protein. Computational prediction tools and conservation analyses suggest that this variant may not impact the protein function. Computational splicing tools suggest that this variant may not impact the RNA splicing. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant is rare in the general population and has been identified in 0/277264 chromosomes by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the pathogenicity of this variant conclusively.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
