This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 10070, where C is replaced by G; at the protein level this means replaces threonine at residue 3357 with arginine — a missense variant. Submitter rationale: Variant summary: BRCA2 c.10070C>G (p.Thr3357Arg) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251130 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.10070C>G has been reported in the literature in breast cancer cells without evidence of a germline origin (e.g. Teng_1996). These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 8640236). Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014, and classified it as likely benign (n=1) or uncertain significance (n=1). Based on the evidence outlined above, the variant was classified as uncertain significance.
