This variant lies in the APC gene (transcript NM_000038.6) at coding-DNA position 3980, where C is replaced by T; at the protein level this means replaces serine at residue 1327 with leucine — a missense variant. Submitter rationale: The p.S1327L variant (also known as c.3980C>T), located in coding exon 15 of the APC gene, results from a C to T substitution at nucleotide position 3980. The serine at codon 1327 is replaced by leucine, an amino acid with dissimilar properties. This variant has been reported in 1/1120 pediatric cancer patients who underwent whole genome sequencing and/or whole exome sequencing; this patient was diagnosed with ependymoma (Zhang J et al. N Engl J Med, 2015 Dec;373:2336-2346). This amino acid position is not well conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Missense alterations in APC are not a common cause of disease (Spier I et al. Genet Med. 2024 Feb;26(2):100992). Based on the available evidence, the clinical significance of this variant remains unclear.
