Submitter rationale: To the best of our knowledge, the APC c.2567G>A (p.R856H) variant has not been reported in individuals with APC-related disease. This variant was observed in 1/18384 chromosomes of the East Asian subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID: 627759). In silico tools suggest the impact of the variant on protein function is inconclusive, though these predictions have not been confirmed by functional studies.
