Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_002294.3(LAMP2):c.959A>G (p.Tyr320Cys), citing Ambry Variant Classification Scheme 2023: The p.Y320C variant (also known as c.959A>G), located in coding exon 8 of the LAMP2 gene, results from an A to G substitution at nucleotide position 959. The tyrosine at codon 320 is replaced by cysteine, an amino acid with highly dissimilar properties. Based on data from gnomAD, the G allele has an overall frequency of <0.01% (2/183450) total alleles studied, with 2 hemizygote(s) observed. The highest observed frequency was 0.01% (2/13857) of East Asian alleles. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
