NM_000416.3(IFNGR1):c.665A>G (p.His222Arg) was classified as Uncertain significance for Disseminated atypical mycobacterial infection by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with arginine, which is basic and polar, at codon 222 of the IFNGR1 protein (p.His222Arg). This variant is present in population databases (rs768805562, gnomAD 0.2%). This variant has not been reported in the literature in individuals affected with IFNGR1-related conditions. ClinVar contains an entry for this variant (Variation ID: 625961). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt IFNGR1 protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change does not substantially affect IFNGR1 function (PMID: 24199198). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
