Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005585.5(SMAD6):c.374C>T (p.Thr125Ile), citing Ambry Variant Classification Scheme 2023: The p.T125I variant (also known as c.374C>T), located in coding exon 1 of the SMAD6 gene, results from a C to T substitution at nucleotide position 374. The threonine at codon 125 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
