This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 6910, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 2304 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q2331X variant in the PLEC gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q2331X variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). We interpret Q2331X as a pathogenic variant.
