Pathogenic — the classification assigned by GeneDx to NM_001271.4(CHD2):c.1234G>T (p.Glu412Ter), citing GeneDx Variant Classification (06012015): The E412X nonsense variant in the CHD2 gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The E412X variant is not observed in large population cohorts (Lek et al., 2016). Although this pathogenic variant has not been reported previously to our knowledge, its presence is consistent with the diagnosis of a CHD2 -related disorder in this individual.
