NM_001013838.3(CARMIL2):c.731C>G (p.Ser244Ter) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The S244X variant in the CARMIL2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The S244X variant is not observed in large population cohorts (Lek et al., 2016). We interpret S244X as a likely pathogenic variant.
