Pathogenic — the classification assigned by GeneDx to NM_000373.4(UMPS):c.670G>T (p.Glu224Ter), citing GeneDx Variant Classification (06012015). This variant lies in the UMPS gene (transcript NM_000373.4) at coding-DNA position 670, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 224 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The E224X variant in the UMPS gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The E224X variant is not observed in large population cohorts (Lek et al., 2016). We interpret E224X as a pathogenic variant.
