Pathogenic — the classification assigned by GeneDx to NM_001142864.4(PIEZO1):c.1105C>T (p.Gln369Ter), citing GeneDx Variant Classification (06012015). This variant lies in the PIEZO1 gene (transcript NM_001142864.4) at coding-DNA position 1105, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 369 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q369X variant in the PIEZO1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q369X variant is not observed in large population cohorts (Lek et al., 2016). We interpret Q369X as a pathogenic variant.
