Pathogenic — the classification assigned by GeneDx to NM_014795.4(ZEB2):c.3243C>A (p.Cys1081Ter), citing GeneDx Variant Classification (06012015): The C1081X variant in the ZEB2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function through protein truncation. The C1081X variant is not observed in large population cohorts (Lek et al., 2016). We interpret C1081X as a pathogenic variant.
