This variant lies in the FRAS1 gene (transcript NM_025074.7) at coding-DNA position 880, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 294 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q294X variant in the FRAS1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q294X variant is not observed in large population cohorts (Lek et al., 2016). We interpret Q294X as a pathogenic variant.
