Pathogenic — the classification assigned by GeneDx to NM_000441.2(SLC26A4):c.301C>T (p.Gln101Ter), citing GeneDx Variant Classification (06012015). This variant lies in the SLC26A4 gene (transcript NM_000441.2) at coding-DNA position 301, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 101 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q101X nonsense variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The variant is not observed in large population cohorts (Lek et al., 2016). The variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. We consider this variant to be pathogenic.
