Pathogenic for KBG syndrome — the classification assigned by Variantyx, Inc. to NM_013275.6(ANKRD11):c.2283C>G (p.Tyr761Ter), citing Variantyx Assertion Criteria 2022: This is a nonsense variant in the ANKRD11 gene (OMIM: 611192). Pathogenic variants in this gene have been associated with autosomal dominant KBG syndrome. This variant likely occurred de novo in the current proband, however, the possibility of parental germline mosaicism cannot be excluded (PS2). This variant introduces a premature termination codon in exon 9 out of 13 and is expected to result in loss of function, which is a known disease mechanism for ANKRD11 in this disorder (PMID: 21782149, 35330407, 28422132) (PVS1). This variant is absent from control populations (https://gnomad.broadinstitute.org/) (PM2). Based on the current evidence, this variant is classified as pathogenic for autosomal dominant KBG syndrome.
