This variant lies in the IL1RAPL1 gene (transcript NM_014271.4) at coding-DNA position 1054, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 352 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The R352X variant in the IL1RAPL1gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The R352X variant is not observed in large population cohorts (Lek et al., 2016). We interpret R352X as a pathogenic variant.
