NM_000308.4(CTSA):c.195G>A (p.Trp65Ter) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The W83X variant has been reported in a patient with galactosialidosis who was homozygous for the W83X variant (Yubero et al., 2016). The W83X variant is not observed in large population cohorts (Lek et al., 2016). The W83X variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. In summary, we interpret this variant as pathogenic.
