This variant lies in the PAX3 gene (transcript NM_181458.4) at coding-DNA position 821, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 274 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The W274X nonsense variant in the PAX3 gene has been reported previously in association with Waardenburg syndrome type 1 (Tassabehji et al., 1995). The variant is not observed in large population cohorts (Lek et al., 2016). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. In summary, we interpret this variant as a pathogenic variant.
