Likely pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000059.4(BRCA2):c.1910-1G>T, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the BRCA2 gene (transcript NM_000059.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1910, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: BRCA2 c.1910-1G>T is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 3 prime acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 237664 control chromosomes (gnomAD). To our knowledge, no occurrence of c.1910-1G>T in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as pathogenic (n=1) and likely pathogenic (n=2). Based on the evidence outlined above, the variant was classified as likely pathogenic.
