Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.4220A>G (p.Tyr1407Cys), citing Ambry Variant Classification Scheme 2023: The p.Y1407C variant (also known as c.4220A>G), located in coding exon 33 of the POLE gene, results from an A to G substitution at nucleotide position 4220. The tyrosine at codon 1407 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
