NM_000179.3(MSH6):c.1093T>C (p.Trp365Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W365R variant (also known as c.1093T>C), located in coding exon 4 of the MSH6 gene, results from a T to C substitution at nucleotide position 1093. The tryptophan at codon 365 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
