Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_032578.4(MYPN):c.2189C>G (p.Thr730Arg), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the MYPN gene (transcript NM_032578.4) at coding-DNA position 2189, where C is replaced by G; at the protein level this means replaces threonine at residue 730 with arginine — a missense variant. Submitter rationale: Variant summary: MYPN c.2189C>G (p.Thr730Arg) results in a non-conservative amino acid change in the encoded protein sequence. Three of four in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251420 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2189C>G has been reported in the literature in individuals affected with Cardiomyopathy (Robyns_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Cardiomyopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. One laboratory classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
