NM_002890.3(RASA1):c.71C>G (p.Ala24Gly) was classified as Likely benign by ARUP Laboratories, Molecular Genetics and Genomics, ARUP Laboratories, citing ARUP Molecular Germline Variant Investigation Process: The RASA1 c.71C>G; p.Ala24Gly variant, to our knowledge, is not reported in the medical literature or gene specific databases. This variant is also absent from the general population databases (1000 Genomes Project, Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. The alanine at codon 24 is weakly conserved and computational algorithms (SIFT, PolyPhen2) predict this variant to be tolerated. Additionally, the majority of pathogenic RASA1 variants are truncating rather than missense (see link). Based on available information, this variant is considered likely benign.
