It is absent from general population databases (1000 Genomes Project, Exome Variant Server, and Genome Aggregation Database), indicating it is not a common polymorphism. The glycine at codon 2107 is highly conserved, and computational algorithms (PolyPhen-2, SIFT) predict that this variant is deleterious. Additionally, another missense variant at this codon (p.Gly2107Ser) and deletion of this amino acid (p.Gly2107del) have been reported in individuals affected with severe hemophilia and are considered pathogenic (see link to FVIII database and references therein).
