Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 8e-06 in 251486 control chromosomes (gnomAD). c.1820G>A has been observed in individuals affected with Inclusion Body Myopathy 2 or GNE myopathy (e.g., Eisenberg_2001, Jiao_2024). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in 10%-<30% of normal enzymatic activity (Penner_2006).
