Uncertain significance for Glycogen storage disease, type II — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000152.5(GAA):c.1381G>A (p.Gly461Ser), citing Genomenon Sequence Variant Interpretation Standards - Updated: GAA p.Gly461Ser (c.1381G>A) is a missense variant that changes the amino acid at codon 461 from Glycine to Serine. To our knowledge, this variant has not been reported in patients affected with a GAA-related disorder in the published literature. At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:18425781). It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify GAA p.Gly461Ser (c.1381G>A) as a variant of uncertain significance.
