NM_001378454.1(ALMS1):c.820A>C (p.Ser274Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S275R variant (also known as c.823A>C), located in coding exon 5 of the ALMS1 gene, results from an A to C substitution at nucleotide position 823. The serine at codon 275 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
