This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 7256, where G is replaced by T; at the protein level this means replaces arginine at residue 2419 with leucine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 2446 of the PLEC protein (p.Arg2446Leu). This variant is present in population databases (rs554586153, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with PLEC-related conditions.
