This variant lies in the TYR gene (transcript NM_000372.5) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1037, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: TYR c.1037-2A>G is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of TYR function. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a canonical 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8e-06 in 250000 control chromosomes (gnomAD). c.1037-2A>G has been reported in the literature in individuals affected with Oculocutaneous Albinism (e.g. Lasseaux_2018, Wei_2022). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
