Submitter rationale: The p.P258T variant (also known as c.772C>A), located in coding exon 8 of the DMD gene, results from a C to A substitution at nucleotide position 772. The proline at codon 258 is replaced by threonine, an amino acid with highly similar properties. Based on data from gnomAD, the A allele has an overall frequency of 0.001466% (3/204652) total alleles studied, with 1 hemizygote observed. The highest observed frequency was 0.01578% (3/19015) of African American alleles. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
