Uncertain significance for Peroxisome biogenesis disorder 7B; Peroxisome biogenesis disorder 7A (Zellweger) — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001127649.3(PEX26):c.725C>T (p.Ser242Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with phenylalanine at codon 242 of the PEX26 protein (p.Ser242Phe). The serine residue is highly conserved and there is a large physicochemical difference between serine and phenylalanine. This variant is present in population databases (rs779828574, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with PEX26-related conditions. ClinVar contains an entry for this variant (Variation ID: 593518). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The phenylalanine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
