Pathogenic for Hajdu-Cheney syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024408.4(NOTCH2):c.5664dup (p.Arg1889fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 593268). This variant has not been reported in the literature in individuals affected with NOTCH2-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Arg1889Thrfs*3) in the NOTCH2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NOTCH2 are known to be pathogenic (PMID: 16773578, 22209762).
